May 11, 2010 (Washington DC) — Both the diabetes drug rosiglitazone (Avandia, GlaxoSmithKline) and the way the FDA has handled concerns about it took another severe battering at a recent US Congressional hearing of the committee that has jurisdiction over the FDA.

The House Appropriations Subcommittee on Agriculture, Rural Development, Food and Drug Administration, and Related Agencies met on April 28 to discuss the report on rosiglitazone released in February by the Committee on Finance of the US Senate.

The report [1], by Sens Max Baucus (D-MT) Charles Grassley (R-IA), concluded that there are "serious health risks associated with Avandia." It also criticized the structure of the FDA, particularly the fact that those who make decisions about drug approvals are the same experts who must later oversee drug safety.

TCF7L2 variants have been associated with type 2 diabetes in multiple ethnic groups.1,2,3,4,5,6,7 Specific associated variants increase risk of type 2 diabetes 1.5-fold in heterozygotes and 2.41-fold in homozygotes, corresponding to a population attributable risk of 21%. This makes TCF7L2 variants the strongest known genetic risk factor for type 2 diabetes.8

TCF7L2 is a transcription factor and key component of the Wnt signaling pathway, and is involved in the development of a wide variety of cell lineages and organs.9 Potential mechanisms by which TCF7L2 variants influence type 2 diabetes include its role in adipogenesis, myogenesis, and pancreatic islet development, as well transcription of the genes for proglucagon and the glucagon-like-peptides GLP-1 and GLP-2, which play a role in post-prandial insulin secretion.10 Additionally, TCF7L2 polymorphisms have been associated with impaired insulin secretion, glucose production, and glucose tolerance via direct effects on pancreatic islet beta-cells.11,12 Thus, while the specific mechanism driving the development of type 2 diabetes remains unclear, there is sufficient evidence to demonstrate that TCF7L2 variants strongly predict the development of type 2 diabetes and/or the progression to diabetes from impaired glucose tolerance.12,13

05/28/09 - Man with HIV convicted of assault with sex -- MCKINNEY, Texas, May 28 (UPI) -- An HIV-positive Texas man faces a long prison term after being convicted of assaulting six women by having unprotected sex with them. Philippe Padieu, 53, a martial arts teacher from Frisco, was convicted Wednesday of aggravated assault, The Dallas Morning News reported. Jurors in Collin County deliberated for about five hours before reaching their verdict.

Maugh II October 25, 2009 * EmailE-mail * printPrint * Share * increase text size decrease text size Text Size Pennsylvania researchers using gene therapy have made significant improvements in vision in 12 patients with a rare inherited visual defect, a finding that suggests it may be possible to produce similar improvements in a much larger number of patients with retinitis pigmentosa and macular degeneration. The team last year reported success with three adult patients, an achievement that was hailed as a major accomplishment for gene therapy. They have now treated an additional nine patients, including five children, and find that the best results are achieved in the youngest patients, whose defective retinal cells have not had time to die off.

05/26/09 - Gum Disease, Heart Disease Share Genetic Link -- TUESDAY, May 26 (HealthDay News) -- A genetic link between gum disease (periodontitis) and heart disease has been discovered by German scientists. "We studied a genetic locus on chromosome 9p21.3 that had previously been identified to be associated with myocardial infarction [heart attack], in a group of 151 patients suffering from the most aggressive, early-onset forms of periodontitis, and a group of 1,097 CHD patients who had already had a heart attack. The genetic variation associated with the clinical pictures of both diseases was identical," Dr. Arne Schaefer said in a European Society of Human Genetics (ESHG) news release.
